Negative staining and Native staining EM
Negative staining, also known as negative contrast staining, is a staining technique used for microscopic examination of opaque liquid specimens. In contrast to traditional dyeing, there is no reaction between the sample and dye solution. So the sample is not colored. A white sample with a low electron density can be contrast with a black background, forming an obvious negative contrast in contrast to density diffirence between the sample and the dye. Negative staining electron microscopy is widely used in sample analysis because it eliminates the needs for complex procedure such as fixation,deny dration, embedding, and ultra-thin slicing.
